Application Note: Vitamin Analysis by HPLC

Column-selection and method-development guidance for water-soluble and fat-soluble vitamin analysis.

This application note provides a method-development framework. Final conditions must be validated with the actual analytes, matrix, detector and laboratory system.

Column selection

Use C30 chemistry for fat-soluble vitamins and structurally related hydrophobic compounds when shape selectivity is valuable. ODS-UG-5 is a practical general-purpose choice for routine reversed-phase vitamin methods. RPAQUEOUS-AR can be evaluated for more polar water-soluble vitamins requiring stronger retention under aqueous conditions.

Sample preparation

Protect the analytical column by clarifying extracts, filtering through a chemically compatible membrane and minimizing particulate or lipid carryover. Light-sensitive vitamins should be protected from unnecessary exposure during preparation and analysis.

Mobile-phase strategy

Begin with a buffered aqueous phase and an organic modifier such as methanol or acetonitrile. Gradient elution is useful when the sample contains vitamins covering a wide polarity range. Select pH and additives according to analyte stability and detector compatibility.

Method checks

Evaluate peak identity, resolution, retention repeatability, recovery, linearity and solution stability. Confirm that the selected column dimensions and particle size are compatible with system pressure and required run time.
