Prepare clean samples
Centrifuge or filter samples using a membrane compatible with the analyte and solvent. Remove precipitates, insoluble excipients and strongly retained matrix components where possible.
Use compatible solvents
Avoid abrupt exposure to immiscible solvents. Change solvent composition gradually and confirm that buffers remain soluble throughout the transition.
Control pressure
Use the recommended flow range, monitor pressure trends and avoid sudden pressure shocks. Persistent pressure increase often indicates contamination in the inlet frit, guard column or packed bed.
Wash after demanding samples
Apply a method-appropriate wash sequence that is strong enough to remove retained material without exceeding phase or hardware compatibility.
Store correctly
Remove buffers before storage and use a suitable storage solvent for the stationary phase. Cap the column securely to prevent evaporation and drying.
